چکیده
Abstract Tomato yellow leaf curl virus (TYLCV) is known to encode 6 canonical viral proteins. Our recent study revealed that TYLCV also encodes some additional small proteins with potential virulence functions. The fifth ORF of in the complementary sense, which we name C5, evolutionarily conserved, but little about its expression and function during infection. Here, confirmed C5 by analyzing promoter activity upstream sequences detecting protein infected cells using a specific custom-made antibody. Ectopic potato X (PVX) vector resulted severe mosaic symptoms higher accumulation levels followed burst reactive oxygen species (ROS) Nicotiana benthamiana plants. was able effectively suppress local systemic post-transcriptional gene silencing (PTGS) induced single-stranded GFP not double-stranded GFP, reversed transcriptional (TGS) GFP. Furthermore, mutation inhibited replication development disease Transgenic overexpression could complement infectious clone encoding dysfunctional C5. Collectively, this reveals pathogenicity determinant RNA suppressor, hence expanding our knowledge functional repertoire proteome.

Natural recombination between Tomato yellow leaf curl virus-is and Tomato leaf curl virus.
The complete genome sequences (2791 and 2793 nt) of isolates of Tomato yellow leaf curl virus-Is (TYLCV-Is) from Spain (SP72/97) and Portugal (Port2/95) were determined. These isolates are closely related to TYLCV-Is isolates reported in Japan (Japan-A and Japan-S) and Israel (Israel/Mild). Comparison of all sequenced isolates of TYLCV-Is showed that part of the genome comprising the intergenic...

Background and Aims: Tomato yellow leaf curl virus (TYLCV) is one of the most destructive viruses of tomato that leads to reduced tomato yield up to 100% in tropical and subtropical regions. In this study, the complete sequence of TYLCV isolate from Hormozgan province, Iran and its recombination evsent was determined. Methods: TYLCV infected tomato was collected from Hormozgan province. Total D...
متن کاملPhenotypic and Molecular Screening of Tomato Germplasm for Resistance to Tomato Yellow Leaf Curl Virus
Tomato yellow leaf curl virus (TYLCV) is a major tomato virus in tropical and subtropical regions. In this study, 134 accessions of Solanum lycopersicum and six accessions of Solanum peruvianum were assessed for resistance to an Iranian isolate of TYLCV. Plants were inoculated using whiteflies (Bemisia tabaci) and the reaction of plants was evaluated based on either disease symptoms or viral DN...

Tomato yellow leaf curl virus (TYLCV) is a supreme pathogen in tropical and subtropical areas. During 2014-2015, a total of 393 tomato samples showing Tomato yellow leaf curl disease (TYLCD) symptoms were collected from six different provinces of Iraq. In serological assays, 55 out of 393 samples (14%) reacted positively with TYLCV-specific antibodies .The presence of TYLCV was verified in 21 (...
متن کاملInteraction with host SGS3 is required for suppression of RNA silencing by tomato yellow leaf curl virus V2 protein.
The V2 protein of tomato yellow leaf curl geminivirus (TYLCV) functions as an RNA-silencing suppressor that counteracts the innate immune response of the host plant. The host-cell target of V2, however, remains unknown. Here we show that V2 interacts directly with SlSGS3, the tomato homolog of the Arabidopsis SGS3 protein (AtSGS3), which is known to be involved in the RNA-silencing pathway. SlS...
